Mechanism of action of colloidal silver
The antibacterial activity of colloidal silver based veterinary products is based on the synergistic effect of colloidal silver particles and benzyldimethyl-[3-myristoylamine)-propyl]ammonium chloride molecules.
Silver particles are slowly oxidized by the oxygen present in the air, releasing silver ions which disrupt operation of transport proteins and enzymes of a pathogen’s electron transport chain (ETC) by penetrating the bacterial cell, interrupting translation and ultimately causing the cell’s death. Slow and controlled oxidation of silver particles is achieved due to them being stabilized with benzyldimethyl-[3-myristoylamine)-propyl]ammonium chloride, which is the key to the prolonged action of colloidal silver based veterinary products.
Benzyldimethyl-[3-myristoylamine)-propyl]ammonium chloride, being a typical cationic surfactant, has its own antibacterial activity – it loosens the cell membrane, disturbing its permeability. At the same time, benzyldimethyl-[3-myristoylamine)-propyl]ammonium chloride helps facilitate the delivery of silver ions inside the cell, enhancing their action.
Due to selective attachment of positively charged silver particles to the negatively charged outer membrane of a bacterial cell which creates high local concentration of silver ions around the cell wall of the pathogen cell, colloidal silver based veterinary products are very effective without any negative side effect on the tissues.
The anti-inflammatory effect of the products is based on the ability of colloidal silver particles to initiate production of anti-inflammatory cytokines and suppress production of proinflammatory mediators.
Colloidal silver initiates a biochemical cascade aimed at reducing inflammation even in areas with no direct contact with silver particles. Selective induction of apoptosis and a decrease in expression and activity of MMP-2 (matrix metalloproteinase-2), MMP-9, proinflammatory cytokines (TGF)-β and (TNF)-α can both be observed in tissues close to the areas treated with colloidal silver but not directly exposed to it. As a result, the processes initiated by colloidal silver affecting the top layers of skin and mucous membranes lead to a systemic decrease in inflammation, which is especially important for treatment of such diseases as bovine endometritis or pet gingivitis.
According to the results of numerous preclinical studies, colloidal silver based veterinary products can be assigned lowest toxicity class. Prolonged use of the product (for more than 10 days) at a dose 10 times higher than the recommended:
did not cause death of laboratory animals;
did not have any negative effect on their general condition;
did not lead to statistically significant changes in the concentration of hemoglobin, the number of erythrocytes, leukocytes, platelets;
did not affect the mass content of total protein, albumin, urea, creatinine in the blood serum of animals;
did not lead to statistically significant changes in the activity of the ALT and AST enzymes.
